Tris(hydroxymethyl)aminomethane (commonly referred to as Tris) is an organic compound with a molecular formula of C4H11NO3 and a CAS Number of 77-86-1. In the pharmaceutical industry, it is called tromethamine (THAM). It is a white crystal or powder. It is a chemical substance soluble in ethanol and water, slightly soluble in ethyl acetate and benzene, insoluble in ether and carbon tetrachloride, corrosive to copper and aluminum, and irritating. Tris is widely used in the preparation of buffers in biochemical and molecular biology experiments. Tris is a weak base with a pKa of 8.1 at 25°C; according to buffer theory, the effective buffering range of Tris buffer is between pH 7.0 and 9.2. The pH of the aqueous solution of Tris base is about 10.5. Generally, hydrochloric acid is added to adjust the pH value to the desired value, and then the buffer solution with the pH value can be obtained. For example, Tris is required in both TAE and TBE buffers (for solubilization of nucleic acids) commonly used in biochemical experiments. Since it contains amino groups, it can undergo condensation reactions with aldehydes. In addition, Tris is also an intermediate in the preparation of surfactants, vulcanization accelerators and some pharmaceuticals. Tris is also used as a titration standard.
